A port is basically an ID number for a network connection, so when your computer receives data, it knows which program to give it to.
As for setting it up, the exact procedure differs between routers, but
this may be of some assistance. Look up your router (skip the big obnoxious advertisement), and find Unreal Tournament. As far as I know, Unreal and Unreal Tournament use the same ports, so the instructions for UT should work for Unreal.
