My monitor's native resolution is 1440p but trying to run NWN 2 at this means the UI is too small to read, so i'm trying to run the game which I've just reinstalled on Win 10, and when I change the resolution to 1080p (with great difficulty, as it locks the whole PC 50% of the time when I do this forcing a hard reset -.-) the mouse pointer ends up being offset to where it's actually "clicking".

Example is on the options screen, hovering the mouse pointer over one square-box option actually ends up selecting the one below.

I have an inkling it's something to do with DPI scaling in Win 10, but I've tried fiddling around with all sorts of options on the NWN 2 exe and nothing seems to work.

Any ideas?

It's a common problem on NWN1, with a simple fix:

Except use a shortcut to this EXE to launch your game:

nwn2main.exe

Then make the same change to the compatibility tab for this EXE as noted in the above post

That particular option doesn't seem to be on my version of Win 10, however it DID prompt me to locate the correct option to tick - it's called "Disable Fullscreen Optimizations"
