When I try to open the game in Fullscreen (or try to change to Fullscreen while in-game) I get the following error message:

An Assertion failed in D:\Dev\chitin\ChVideo.cpp at line number 4256
Programmer says: No valid video modes supported

I can play in windowed mode just fine, but I'd really rather not. I'm not using any mods and I've tried reinstalling the game.

I'm using Windows 10.

Baldur's Gate is a fixed 4:3 asset game. Your resolution is a non-standard 16:9 resolution. Try temporarily changing your desktop resolution to a standard one and see if that helps.

Thank you so much! My monitor doesn't have too wide a range of resolutions available, but I connected it to my TV and it all just works, so it was definitely a resolution issue.

Thank you!

Excellent. Have fun.
You can also try to look at the graphic card settings, some have the option to keep the horizontal (and vertical as well) aspect ratio when you launch games that use the 4:3 one (like Baldur's Gate). That way you can play fullscreen with two black bars to the side and keep the correct aspect ratio. I haven't tested it on Windows 10, though.

That won't work. The error (in this case)

"An Assertion failed in D:\Dev\chitin\ChVideo.cpp at line number 4256
Programmer says: No valid video modes supported "

is caused by a non-standard desktop resolution, further exacerbated by the monitor not having " too wide a range of resolutions available".

That's weird, I have laptop at my home that has 16:9 aspect ratio of the screen (resolution - 1366 x 768) and there's no such error. Is this problem Windows 10 exclusive, or does it have something to do with certain larger monitors?

I have no idea. I don't have Windows 10. It's possible, though. The physical size of the monitor doesn't factor at all.
