It varies with every mod, and the creators usually state in the mod's readme file if you need to start a new game or not for the changes to take effect, so it's always best to read that when you're in doubt.
And, as I know to my cost, do not install or change the widescreen mod mid game!

The answer is 'it depends'. It depends on what the tweak pack alters, but usually a banter pack or NPC will fit in without issues, other than some content may be missed. As has already been said, check the readme files, and never install or uninstall the widescreen mod mid game... ever!
