Hi, if you mean the door in the, let's say, top right corner of the map, then you need to kill Salacious Vincenzio first to get the key for this door. You can find Vincenzio in a room at the bottom of the map.
These two doors on level 2 can not be opened at all. The key You obtained from the necromancer actually fits to the door at level 1 near the entrance to the surface. That gets You to level 3, where You must get the key from Salacious Vincenzio as Kovi said, then open the door at north of that map and ascend up to level 2 from the other side. The 2 doors remain closed forever.

The OP certainly means level 3 doors (which is the second level available and only one with 2 blocked unclickable doors). As he probably figured it out by now I won't bother explaining in detail, only short notes (for other folks):

lvl 1 -> lvl 3 (key) -> lvl 1 -> lvl 2 -> key (for northernmost door) -> lvl 3 (other side of blocked doors, key) -> lvl 2 -> lvl 4
Path is designed to be linear. You can see on which level you are by looking at your savegame details on that level.

What I want to know is are the two blocked doors on level 3 really blocked forever? No hidden levers or wall switches?
Need the info for my new complete Act 3 map guide I am working on. Also, nice touch with that 10k hit point wooden barrel on level 3, lol.

Yes, as far as I know, those two doors are blocked for good.
