I'm on Win XP, and I've been able to run TIM3 before with no problems, and there's no reason (At least that I can think of) as to why TIM3 has suddenly stopped working.

I load it up, it gets to the Sierra splash screen and then a Windows error pops up. Is there any way I can find out what's causing it to not work?

What does the Windows error message say exactly?

Go to the directory the game was installed to, C:\Program Files\GOG.com\The Incredible Machine Series\The Incredible Machine 3\ by default, right click TIMWIN.EXE click properties and go to the compatibility tab and make sure "run this program in compatibility mode for" is checked and "Windows 95" is selected.

You could try temporarily disabling your antivirus and running the game, I suggest disconnecting from the internet before you do this.

The error message was one of those generic Windows 'This Application Has Encountered An Issue And Needs To Close' type errors. Bit annoying, really :p
